摘要(Abstract):

【摘要】目的 探讨Let-7a和miR-96分子在胰腺癌中对KRAS基因的抑制作用及其致病机理。方法 检测胰腺癌细胞系中miRNA分子(Let-7a和miR-96)水平、K-RAS基因的mRNA和蛋白质水平来分析Let-7a和miR-96水平与K-RAS蛋白表达之间的关系,并且升高胰腺癌细胞中Let-7a和miR-96水平,检测K-RAS蛋白的改变情况。结果 1)胰腺癌细胞系中K-RAS的mRNA保持在一个较高的水平,并且K-RAS的蛋白质表达也维持在一个相对较高的水平;2)胰腺癌细胞系中的miRNA分子--Let-7a和miR-96水平均较低;3)升高胰腺癌细胞中Let-7a或miR-96水平可降低K-RAS的蛋白质表达水平。结论 胰腺癌中的Let-7a和miR-96水平有一定程度的降低,正常条件下它们通过调控K-RAS基因来发挥对肿瘤的抑制作用,而在肿瘤中这种改变削弱了对K-RAS的抑制作用,从而参与了肿瘤的致病过程。
【Abstract】Objective To investigate the inhibition to KRAS by Let-7a and miR-96 molecules in pancreatic cancer. Methods The level of KRAS mRNA and protein were detected in 9 pancreatic cancer cell lines, then in 7 of these cell lines, the abundance of Let-7a and miR-96 were also determined. At last, the level of Let-7a or miR-96 were increased in PANC-1 cell line by transfection, then the change of KRAS protein was observed. Results A high level of KRAS mRNA were observed in 9 pancreatic cancer cell lines, and KRAS protein is also in a relatively high level. But Let-7a and miR-96 maintained a low level in them. Increase of Let-7a or miR-96 decreased the expression of KRAS in PANC-1 cell line. Conclusion The level of Let-7a and miR-96 have decreased to some content in pancreatic cancer compared to normal condition, and this attenuation of inhibition to KRAS by them contributed to pancreatic cancer formation and progression.
